Because of the increasing resistantce to antifungal agents, determination of an efficient strategic plan for treatment of fungal diseases is an important issue in clinical mycology. Material and Methods:Many methods have been introduced and developed to determine invitro susceptibility tests. During the recent years, flow cytometry has been developed to solve the problem, and many papers have documented the usefulness of this technique. This study was done to evaluate the invitro susceptibility of standard PTCC strain and some clinical isolates of Candidaevaluated by macrodilution broth method according to NCCLS guidelines and flow cytometry susceptibility test. This descriptive study was done in Iran Medical University. Clinical isolates of Candidawere treated by Amphotericin B, Clotrimazole, Fluconazol, Ketoconazol and miconazole and then evaluated by flow cytometry and microdilution. Data was gathered in a questionnaire and analyzed by descriptive statistics. Results:The data indicated that macrodilution broth methods and flow cytometry have the same results in determination of MIC for amphotericinB, clotrimazole, fluconazole, ketoconazole and miconazole in Candida albicans PTCC5027 as well as clinical candida isolate, such as C.albicans, C.kefyr, C.glabrata, C.parapsilosis and C.dubliniensis. Conclusion:Comparing the result obtained by macrodilution broth and flow cytometry methods revealed that flow cytometry was more convenient and faster than broth methods. It is suggested that flow cytometry susceptibility test can be used as a powerful tool in determination of MIC and administration of the best antifungal drug in treatment of patients with Candida infections especially systemic candidiasis.}, keywords = {Antifungal Drugs,Flow cytometry,Susceptibility89}, title_fa = {اثرات داروهای ضد قارچی متداول در نمونه های بالینی انواع مختلف کاندیدا آلبیکنس}, abstract_fa = {مقدمه: در چند دهه اخیر، میزان وقوع عفونتهای قارچی در بسیاری از کشورها افزایش یافته است. به دلیل پیدایش مقاومت به عوامل ضد قارچی تعیین طرح استراتژیک مؤثربرای درمان بیماریهای قارچی یک موضوع مهم در قارچشناسی بالینی است. روش کار: روشهای زیادی برای تعیین تست حساسیت آزمایشگاهی معرفی شدهاند. اخیراً، فلوسیتومتری برای حل مشکل معرفی شده است. مقالات زیادی به مفید بودن این تکنیک استناد کردهاند.این مطالعه با هدف بررسی حساسیت آزمایشگاهی یک سوش استاندارد PTCC و تعدادی ایزولههای بالینی از کاندیدا مطابق با دستورالعمل NCCLS با روش ماکرودیلوشن براث و تست حساسیت فلوسیتومتری انجام شده است. این مطالعه توصیفی بر روی قارچ ها در دانشگاه علوم پزشکی ایران انجام شد. نمونه های بالینی کاندیدایی جدا شده تحت اثر دارهای ضدقارچی مانند آمفوتریسینB، فلوکونازول، کلوتریمازول، کتوکونازول، میکونازول به روش فلوسیتومتری و ماکرودیلوشن قرار گرفتند. مشخصات نمونه ها، نتایج آزمایشات در پرسشنامه و برگه مشاهده جمع آوری و سپس با استفاده از آمار توصیفی و جداول توزیع فراوانی پردازش شد. نتایج: دادهها نشان دادند که روش ماکرودیلوشن براث و فلوسیتومتری نتایج مشابه در تعیین MIC برای آمفوتریسین B، کتوکونازول، کلوتریمازول، فلوکونازول و میکونازول داشتند. نتیجه گیری: مقایسه نتایج حاصله با روش ماکرودیلوشن براث و فلوسیتومتری در مورد کاندیدا آلبیکنس PTCC5027 و همین طور ایزولههای بالینی نشان داد که فلوسیتومتری خیلی راحتتر و سریعتر از روشهای براث است.پیشنهاد میشود که تست حساسیت فلوسیتومتری بهعنوان یک ابزار قوی در تعیین MIC و تجویز بهترین داروی ضد قارچی در درمان بیماران با عفونتهای کاندیدائی به خصوص کاندیدیازیس سیستمیک استفاده شود}, keywords_fa = {داروهای ضد قارچی,فلوسیتومتری,روش ماکرودیلوشن}, url = {https://mjms.mums.ac.ir/article_5599.html}, eprint = {https://mjms.mums.ac.ir/article_5599_a9c2db87cda2e5cfe0f9f93b708a9ac2.pdf} }
