تأثیر یک دوره تمرین استقامتی همراه با دریافت مکمل سماق بر سطوح سرمی سیرتوئین‌های میتوکندریایی (سیرتوئین ۳ و ۴) و آدنوزین مونوفسفات کیناز موش‌های ویستار نر آلزایمری

چکیده
مقدمه: هدف این مطالعه بررسی تأثیر ۱۲ هفته تمرین استقامتی همراه با دریافت سماق بر سطح سرمی SIR3، SIR4 و AMPK در موش‌های نر آلزایمری بود.
روش‌ کار: در این مطالعه تجربی، ۳۵ سر رت نژاد ویستار بـا سـن ۱۰-۸ هفته و میـانگین وزنـی ۳۰±۲۳۰ گرم به‌صورت تصادفی به پنج گروه کنترل، آلزایمری، آلزایمری همراه با دریافت مکمل سماق، آلزایمری همراه با تمرین استقامتی و آلزایمری همراه با تمرین استقامتی و دریافت مکمل سماق تقسیم شدند. برای القای آلزایمر، ۸ میلی‌گرم/کیلوگرم تری متیل تین کلراید به همراه ۲۰۰ میکرولیتر نرمال سالین به موش‌ها تزریق شد. تمرین استقامتی به مدت ۱۲ هفته و ۵ روز در هفته در استخر ویژه موش انجام گرفت. ۴۸ ساعت بعد از آخرین جلسه تمرین، نمونه خونی گرفته شد. سطح سرمی SIR3، SIR4 و AMPK به روش الایزا مورد ارزیابی قرار گرفتند. داده‌ها با روش آنوای یک‌سویه به همراه آزمون تعقیبی توکی در سطح معنی‌داری ۰۵/۰P< بررسی شد.
نتایج: نتایج تحقیق حاضر نشان داد که ۱۲ هفته تمرین استقامتی موجب افزایش میزان SIR3(002/0P<)، SIR4 (001/0P<) و AMPK (031/0P<) نسبت به گروه آلزایمری شد. بعلاوه، اضافه‌کردن سماق به برنامه تمرین استقامتی اثر معنی‌داری بر میزان SIR3(001/0P<)، SIR4 (001/0P<) و AMPK (0001/0P<) داشت.
نتیجه‌گیری: نتایج ما پیشنهاد می‌کند که تمرین استقامتی با بهبود سطح سیرتوئین ۳، سیرتوئین ۴ و آدنوزین مونوفسفات کیناز در موش‌های آلزایمری همراه است و دریافت ترکیب آنتی‌اکسیدانی سماق منجر به مضاعف شدن این پاسخ‌ها می شود.

Introduction: The aim of this study was to investigate the effect of 12 weeks of endurance training with sumac intake on the serum levels of SIR3, SIR4 and AMPK in Alzheimer's male rats.
Methods: In this experimental study, 35 Wistar rats aged 8-10 weeks and average weight of 230±30 grams were randomly divided into five groups: control, Alzheimer's disease, Alzheimer's disease with sumac supplementation, Alzheimer's disease with endurance training, and Alzheimer's disease with They were divided by endurance training and receiving sumac supplements. To induce Alzheimer's disease, 8 mg/kg of trimethyl tin chloride along with 200 microliters of normal saline were injected into mice. Endurance training was done for 12 weeks and 5 days a week and blood samples were taken. Serum levels of SIR3, SIR4 and AMPK were evaluated by ELISA method. The data were analyzed by one-way ANOVA method along with Tukey's post hoc test at a significance level of P<0.05.
Results: The results of this research showed that 12 weeks of endurance training increased the levels of SIR3 (P<0.002), SIR4 (P<0.001) and AMPK (P<0.031) compared to the Alzheimer group. In addition, adding sumac to the endurance training program had a significant effect on SIR3 (P<0.001), SIR4 (P<0.001) and AMPK (P<0.0001).
Conclusion: Our results suggest that endurance training is associated with improving the levels of sirtuin 3, sirtuin 4 and adenosine monophosphate kinase in Alzheimer's rats, and receiving sumac antioxidant compound leads to doubling of these responses.
