پلاسمید رپلیکون تایپینگ جدایه های اشریشیاکلی چسبنده ی روده ای جدا شده از کودکان اسهالی با استفاده از روش PCR مبتنی بر تکثیر گروه های ناسازگار

چکیده

سابقه و هدف: پلاسمید ها در انتقال افقی ژن ها به ویژه مقاومت آنتی بیوتیکی و عوامل بیماری زا نقش حیاتی دارند و مطالعات چندانی روی نوع پلاسمیدهای حمل شده توسط پاتوتیپ های اشریشیاکلی چسبنده ی روده ای انجام نگرفته است. هدف اصلی این مطالعه دسته بندی پلاسمید های جدایه های اشریشیاکلی چسبنده ی روده ای جدا شده از کودکان اسهالی بر اساس گروه های ناسازگار عمده پلاسمیدی به روش Johnsnonو همکاران(2007) می باشد.
روش بررسی: این مطالعه توصیفی بر روی 44 جدایه اشریشیاکلی چسبنده ی روده ای جدا شده از نمونه مدفوع کودکان اسهالی به منظور شناسایی 18 رپلیکون پلاسمیدی به روش Johnsnonو همکاران(2007) انجام گرفت.
یافته ها: نتاج نشان داد که رپلیکون پلاسمید FreP با (54/79%)35 بیشترین فراوانی و رپلیکون پلاسمید L/M با (27/2%)1 کمترین فراوانی را داشت. رپلیکون IncFIC ,IncN, IncW, Inc X, Inc HI1, Inc HI2 و IncFIIA در هیچکدام از جدایه ها یافت نشد.
نتیجه گیری: نتایج این مطالعه بیانگر تنوع بالای پروفایل گروه های ناسازگار پلاسمیدی در بین جدایه های مورد مطالعه بود.

Introduction: Plasmids play a the crucial role in the horizontal transfer of genes, especially antibiotic resistance and pathogenic agents, and not epidemiological studies have been conducted on the type of plasmids transmitted by Enteroaggregative Escherichia coli pathotypes. The main purpose of this study is to classify the plasmids of Enteroaggregative Escherichia coli isolates isolated from children with diarrhea based on the major incompatible plasmid groups according method of Johnsnon et al(2007).
Materials and Method: This descriptive study was performed on 44 isolates of Enteroaggregative Escherichia coli Isolates Isolated from Diarrheal to identify 18 plasmid replicons by The method of Johnsnon et al(2007).
Results: The results showed the FreP 35(79.54%) plasmid replicon was the most frequent and L/M plasmid replicon was the least frequent with 1(2.27%). IncFIC, IncN, IncW, Inc X, Inc HI1, Inc HI2 and IncFIIA replicons were not observed in any of the isolates.
Conclusion: The results of this study indicated a high diversity of profile plasmid incompatibility groups among the studied isolates.
