اثر 12 هفته تمرین مقاومتی بر بیان ژن Mafa در بافت پانکراس و انسولین سرم در رت های دیابتی نوع 2

چکیده

مقدمه: افزایش بیان Mafa  با افزایش ترشح انسولین از سلول‌های بتای پانکراس و پیامد آن بهبود در نیم‌رخ گلیسمیک در بیماران دیابتی نوع دو است. این مطالعه با هدف تعیین اثر 12 هفته تمرین مقاومتی بر بیان ژن Mafa در بافت  پانکراس، گلوکز، انسولین در رت های دیابتی نوع 2 انجام گرفت.
روش کار: جامعه آماری را همه رت های نر ویستار انستیتو پاستور ایران تشکیل می دهند که از بین آنها 14 سر رت نر 10 هفته ای در دامنه وزن 20 ± 220 گرم انتخاب شدند و توسط تزریق درون صفاقی نیکوتین آمید + STZ دیابتی نوع 2 شدند. سپس به شیوه تصادفی به گروه های مقاومتی (12 هفته، 5 جلسه در هفته، 7 = n) و کنترل (7 = n) تقسیم شدند. سطوح ناشتایی گلوکز، انسولین سرم و بیان ژن Mafa در سلول های بتای پانکراس دو گروه بعد از آخرین جلسه تمرینی اندازه گیری و توسط آزمون تی مستقل بین دو گروه مقایسه شدند.
 نتایج: تمرین مقاومتی به کاهش معنی دار گلوکز ناشتا  و افزایش معنادار سطوح انسولین سرم در مقایسه با گروه کنترل منجرشد. تمرینات مقاومتی همچنین به افزایش بیان Mafa در بافت پانکراس منجر شد) 0001/0 > p).
نتیجه گیری: احتمالا افزایش سطوح انسولین سرم در پاسخ به تمرینات مقاومتی در رت های دیابتی نوع 2 را می توان به افزایش بیان Mafa در بافت  پانکراس نسبت داد.

Abstract
Introduction: Increased Mafa expression with increased insulin secretion from pancreatic beta cells and consequent improvement in glycemic profile in type 2 diabetic patients. This study aimed to determine the effect of resistance on Mafa expression in pancreas tissue, glucose and insulin in type 2 diabetes rats.
Material and Methods: The statistical population consisted of all male Wistar rats of the pastor inistitue of Iran, among which 14 were male rat 10 weeks old weighting 220 ± 20 g. Type 2 diabetes induced by Intraperitoneal injection of nicotinamid + STZ. Then rats divided randomly into resistance (6 weeks, 5 times/weekly, n=7) and control (n=7) groups. Fasting glucose, serum insulin and Mafa expression in Pancreas tissue of both groups were measured after lasted exercise and compared between 2 groups by independent T test.
Results: Resistance training significantly reduced fasting glucose and significantly increased serum insulin levels compared with the control group. Resistance training also increased Mafa expression in pancreatic tissue( p>0/0001).
Conclusion: Based on these data, increased serum insulin in response to resistance training in T2D rats may be attributed with increased Mafa expression in pancreas tissue.
