بررسی تغییرات برخی نوروتروفین ها در بافت هیپوکامپ در یک مدل انسفالومیلیت خود ایمن تجربی متعاقب تمرین استقامتی و ژل رویال

چکیده

مقدمه: اختلالات نروتروفینی مکانیسمی شایع در بیماری مولتیپل اسکلوروزیس می باشد. با توجه به نقش اساسی نروتروفین ها در عملکرد شناختی، مطالعه حاضر با هدف بررسی تغییرات برخی نوروتروفین ها در بافت هیپوکامپ در یک مدل انسفالومیلیت خود ایمن تجربی (EAE) متعاقب تمرین استقامتی (ET) و ژل رویال (RJ) انجام شد.
روش‌ها: در این مطالعه تجربی 49 سر موش صحرایی ماده نژاد اسپراگو-داولی مبتلا به EAE (توسط ادجوانت فروند کامل) به گروه‌های (1) EAE، (2) شم (Sh)، (3) RJ mg/kg 50 (RJ50)، (4) RJ100، (5) ET، (6) ET+RJ50، (7) ET+RJ100 تقسیم شدند، برای بررسی اثر EAE بر متغیر‌ها تعداد 7 سر موش صحرایی سالم در گروه کنترل سالم (HC) قرار گرفتند. ET به مدت پنج هفته، چهار جلسه در هفته با سرعت 11-15 متر بر دقیقه به مدت 30 دقیقه انجام شد و RJ با دوزهای (mg/kg/day 50 و 100) به صورت صفاقی تزریق گردید. برای تجزیه و تحلیل یافته‌ها از آزمون آنالیز واریانس یک-راهه و آزمون تعقیبی توکی استفاده شد(05/0≥P).
نتایج: نتایج مطالعه حاضر نشان داد مقادیر BDNF در گروه ET به طور معنی داری بیشتر از گروه EAE بود. نتایج مطالعه حاضر نشان داد BDNF در گروه های RJ50، RJ100، های ET+RJ50 و ER+RJ100 به طور معنی داری بیشتر از گروه EAE بود. علاوه بر این مقادیر بیان ژنی TrKB در گروه RJ100 به طور معنی داری بیشتر از گروه RJ50 بود. همچنین TrKB در گروه های RJ50، RJ100، های ET+RJ50 و ER+RJ100 به طور معنی داری بیشتر از گروه EAE بود.

Introduction: Neurotrophin disorders are a common mechanism in multiple sclerosis. Given the essential role of neurotrophins in cognitive function, the present study aimed to investigate the changes in some neurotrophins in hippocampal tissue in an experimental autoimmune encephalomyelitis (EAE) model following endurance exercise (ET) and royal jelly (RJ).
Methods: In this experimental study, 49 female Sprague-Dawley rats with EAE (with complete Freund's adjuvant) were divided into groups: (1) EAE, (2) Sham (Sh), (3) RJ mg/kg 50 (RJ50), (4) RJ100, (5) ET, (6) ET+RJ50, (7) ET+RJ100. To investigate the effect of EAE on the variables, 7 healthy rats were placed in the healthy control (HC) group. ET was performed for five weeks, four sessions per week at a speed of 11-15 m/min for 30 minutes, and RJ was injected intraperitoneally at doses of (50 and 100 mg/kg/day). One-way ANOVA and Tukey's post hoc test were used to analyze the findings (P≥0.05).
Results: The results of the present study showed that BDNF levels in the ET group were significantly higher than in the EAE group. The results of the present study showed that BDNF levels in the RJ50, RJ100, ET+RJ50, and ER+RJ100 groups were significantly higher than in the EAE group. In addition, TrKB gene expression levels in the RJ100 group were significantly higher than in the RJ50 group. Also, TrKB levels in the RJ50, RJ100, ET+RJ50, and ER+RJ100 groups were significantly higher than in the EAE group.
