Background: This study examined the effects of high-intensity aerobic training and myo-inositol supplementation on the expression of UCP1, PGC-1α, and PRDM16 genes associated with adipose tissue browning in obese rats.
Methods: In this experimental study, 50 male Wistar rats were assigned after high-fat-diet-induced obesity to five groups of 10: healthy control, obese control, high-intensity aerobic training, myo-inositol, and high-intensity aerobic training plus myo-inositol. Interventions were performed for 8 weeks. Relative expression of UCP1, PGC-1α, and PRDM16 in adipose tissue was assessed by Real-Time PCR using the 2^-ΔΔCt method. Data were analyzed using one-way analysis of variance, Tukey’s post hoc test, and two-way analysis of variance.
Results: Obesity reduced UCP1, PGC-1α, and PRDM16 gene expression and increased visceral adipose tissue weight. Both high-intensity aerobic training and myo-inositol increased expression of these genes; however, the greatest increase was observed in the combined-intervention group. The training × myo-inositol interaction was significant for all three genes.
Conclusion: Combining high-intensity aerobic training with myo-inositol supplementation may enhance gene-expression responses associated with adipose tissue browning in an animal model of obesity.
