تأثیر هشت هفته تمرین هوازی بر سطوح پروتئین‌های PPAR-α SREBP1c, و CHREBP کبدی در رت‌های دارای دیابت نوع دو

چکیده

مقدمه: مقاومت به انسولین (IR) از مهمترین عوامل ایجادکننده اختلالات کبدی مشخص شده و با کاهش بیان گیرندۀ فعال‌شده با تکثیر پرواکسیزوم آلفا (PPAR-α) ، افزایش بیان پروتئین متصل‌شونده به عنصر تنظیمی استرول (SREBP-1C) و پروتئین متصل‌شونده به عنصر پاسخ‌دهنده کربوهیدرات (ChREBP)کبدی همراه است. هدف از مطالعه حاضر بررسی اثر هشت هفته تمرین هوازی بر سطوح پروتئین‌های PPAR-α ، SREBP-1C و ChREBP موش‌های دیابتی نوع دو می‌باشد.
روش کار: در این مطالعه تجربی ، 27 سر موش نر ویستار، به صورت تصادفی به سه گروه کنترل سالم، کنترل دیابتی و دیابتی تمرین تقسیم شدند. تمرینات هوازی با شدت متوسط و پیش‌رونده به مدت 8 هفته و هر هفته پنج جلسه روی تردمیل انجام شد. پس از هشت هفته تمرین، میزان بیان پروتئینPPAR-α ، SREBP-1cو ChREBP در بافت کبد به روش الایزا و میزانIR با استفاده از فرمول HOMA-IR اندازه‌گیری شد. برای مقایسه بین گروه‌ها از آزمون آنالیز واریانس یک طرفه و آزمون تعقیبی توکی استفاده شد و داده‌ها در سطح 05/0 p< آزمایش شدند.
نتایج: نتایج آزمون تعقیبی کاهش معنادار IR در بین گروه‌ تمربن در مقابسه با گروه دیابت کنترل را نشان داد(005/0 p<). بین گروه‌ها تغییر معتاداری در سطح پروتئین‌های PPAR-α ، SREBP-1C و ChREBP مشاهده شد(005/0 p<).
نتیجه‌گیری: تمرینات هوازی به مدت هشت هفته منجر به بهبود مقاومت به انسولین در موش‌های دیابتی توع دو شد. تفاوت معتاداری در بیان پروتئین‌های کبدی در بین گروه‌ها مشاهده شد.

چکیده [English]

Introduction: Insulin resistance (IR) is known as the most important cause of Liver Disorders, Which is associated with decreased expression of Peroxisome proliferator-Activated Receptor-alpha (PPAR-α) and increased expression of Sterol regulatory Element Binding Proteins (SREBP-1C) and Carbohydrate response element binding protein (CHREBP) in diabetic type 2 male rats. The aim of this study was to evaluate the effect of eight weeks of aerobic exercise on hepatic PPAR-α , SREBP-1C and CHREBP expression and IR in n diabetic type 2 rat.
Materials and methods: In this experimental study, 27 male Wistar mice were randomly divided into three groups of healthy control, diabetic control and diabetic exercise. Medium and progressive aerobic exercise protocol on the treadmill was performed and The trainings lasted 8 weeks and 5 days/week. At the end of the trainings PPAR-α, SREBP-1C and CHREBP expressions were examined via ELISA technique in the liver tissue. One-way Analysis of variance (ANOVA) test were used to compare the groups and the significance level was considered at p < 0.05.
Results: The post-hoc test results indicated a significant reduction in IR between the diabetic control group and aerobic training (P<0.05). Despite a notable increase in hepatic expression, it PPAR-α, SREBP-1C and CHREBP was statistically significant
Conclusion: aerobic exercise for eight weeks improved IR in type 2 diabetic rats. There was a significant difference in the expression of liver proteins between the groups.
